The 50,000 population threshold applies to the larger community served by institutions. For universities, this means more than their immediate enrolled students, and with a population of about 1.1 million people in the Buffalo–Niagara Falls metropolitan area, the University of Buffalo would qualify for the ADA requirement.
Now, the way they are doing the compliance rollout may not be the best. For example, it's possible to grandfather content that wasn't previously required to be accessible behind a password. Their timeline also seems pretty tight, if in the Spring of 2025 you weren't required to submit accessible material for your class, with a deadline set in April 2026.
But the ADA population requirement isn't in question in this case.

Some rather stunning stencilling on the walls of Alexander 'Greek' Thomson's 1850s Holmwood House on the Southside of Glasgow. If I remember correctly, this was part of the design by Thomson himself uncovered during restoration work. This involved carefully scraping off the layers of paint and wall paper which had been added to the interior over the century and a half or so since it was constructed to reveal as much of the original details as possible.
